Doctor Octopus: Defeat Doctor Octopus as a boss in Story mode. For Freeplay mode you must purchase him. 

Other characters can be purchased at the shop for the indicated number of tokens.
Blade: 1,000 tokens. 
Doctor Octopus: 500 tokens. 
Green Goblin: 750 tokens. 
Sandman: 1,250 tokens. 
Venom: 1,500 tokens.
