Play as Gaara:
Complete Story mode under the Hard difficulty setting.

Play as Gai:
Complete Story mode without using any continues.

Play as Itachi:
Complete Arcade mode with default characters.

Play as Jiraiya:
Successfully complete Arcade mode as Tsunade.

Play as Kyubi Naruto:
Complete Arcade mode with Itachi.

Play as Orochimaru:
Successfully complete Arcade mode as Jiraiya.

Play as Sasuke Cursed Seal 2:
Unlock all other characters, then complete the Arcade mode as Sasuke again.

Play as Temari:
Complete Story mode under the Normal difficulty setting.

Play as Tsunade:
Complete one of the following tasks to unlock Tsunade.

Unlock Gai, Temari, and Gaara. This can be done by completing Story mode under the Normal or Hard difficulty setting without using any continues.
Successfully complete Story mode under the Difficult setting using no continues.
